I'll pick Barney Fife and Floyd from The Andy Griffith Show, followed by Mr. Carlin from The Bob Newhart Show, and Fred Mertz from I Love Lucy. Also, Jimmy Olsen, Perry White and Lois Lane from The Adventures of Superman, Mr. Haney from Green Acres, Colonel Blake from M*A*S*H (could add several more from that series), Lou Grant from The Mary Tyler Moore Show, Buddy Sorrell from The Dick Van Dyke Show, Gale Gordon from Our Miss Brooks and The Lucy Show, Maynard G. Krebs from Dobie Gillis, Paul Drake from Perry Mason, Inspector Queen from Ellery Queen, Festus and Doc Adams from Gunsmoke, Chief Clifford from McCloud, etc.
There's a bunch more I could pick, like Mr. Spock from Star Trek, though he essentially became a second lead very quickly, so I disqualify him. I've watched very few commercial TV shows these past few decades, so my list is limited to the older stuff.

In terms of shows that I saw on first-run I'd go with Roz Doyle in Frasier for character development and Jeeves from Fresh Prince of Bel-Air for change of pace. I'd give a special mention to Bernard Woolley from Yes Minister.

Jim mentioned my all time favorite two. Festus and Doc. But I will add a few more. Grady on Sanford and Son, Alice Kramden on the Honeymooners. (Audrey Meadows was a gem.) Eddie Haskell on Leave it to Beaver. Cliff Clavin on Cheers. Baldric in Black Adder.

I have two complete sets, one DVD and one BluRay. As much as I love Harris he took the show into too much comic land. If you look at the unaired pilot without him it is a much more realistic show with realistic problems.
